Research & Development at Maine Biotechnology Services has designed a novel method for purification of fusion product supernatants at the fusion screening phase of hybridoma development. At the most critical juncture in a successful antibody development project, the customer is able to gather more data efficiently and affordably for selection of the most suitable clone. By providing quality purified antibody during the fusion phase, MBS allows our customers unprecedented insight and access to data including specificity, relative affinity and other physical characteristics. A second plate of complementary biotin conjugates can then be generated giving the ability to test matched pairing capabilities. The additional data allows better decision making for subcloning, production, purification and final selection of the antibody to be used in the appropriate application. The new service, MultiPure, has surpassed previous offerings by allowing a high throughput of samples in a 96 well format, exceeding our former block purification method which had a 12 sample capacity.
Application
MultiPure is a proprietary process developed by the R&D lab at Maine Biotechnology Services to give customers the best chance at attaining precisely the monoclonal antibody that will fit their needs by providing purified antibody during the fusion phase of the hybridoma development process; allowing data to drive clone selection.

Technology
Fusion products are expanded and supernatant is collected in up to 15mL quantities. The antibody is bound to a resin and eluted into a sealed 96 well plate. The typical yield for the process is 500ug-1mg. The range of concentrations seen with the MultiPure process is 0.5mg/mL-2.0mg/mL. Once purified, the antibody can be sterile filtered. Customers may also choose to add Sodium Azide. Custom buffers are available by customer request, with minimal loss of material and no measurable impact on the antibody purity by SEC.
